Just read a brilliant piece of advice on Reddit. If you have a player playing on a Thursday night and you currently have him in your flex spot. Move that player to the RB/WR position and keep your Flex spot available.

If you use the flex early (say Eddie Lacy since he plays tonight) and one of your other RB get sick/hurt/etc and your next best player isn't a replacement level RB (say Roy Helu), someone you wouldn't want to put into the RB2 slot on your lineup, but you are forced to play him at RB2 because the flex position has been used. If you leave the flex open you can start a WR or TE in your flex spot and not be punished.

TLDR: Try to keep your flex position open as long as possible
